    A method that uses a next-generation sequencer to simultaneously determine the base sequences of a huge number of DNA fragments.

Acceptable days are Monday to Thursday.
Please avoid duplicate requests with other items.
NGS method will be used to determine numerical or structural abnormalities in chromosomes in TE cells. The types to be determined are euploid embryos, aneuploid embryos, and mosaic embryos.
The influence of contamination is greater with this testing method, so please be careful when collecting specimens. Please see below for specimen collection methods.

-About PGT-A〔NGS method〕
This test can only be requested from medical institutions approved by the Japan Society of Obstetrics and Gynecology as PGT-A implementation facilities. In addition, a contract must be made in advance regarding the request for this test. When requesting this test, please comply with the position on preimplantation genetic testing for infertility and recurrent pregnancy loss.

-Result judgement
The test results will be graded from A to D based on the following criteria.
A:Embryos whose autosomes are euploid
B:Embryos that are mosaic between cells with numerical or structural abnormalities of autosomes and euploid cells of autosomes
C:Embryos with autosomal aneuploidies or structural abnormalities
D:Embryos whose analysis results cannot be determined (which do not meet the quality standards)

-Subject patients
This test is in accordance with the Japan Society of Obstetrics and Gynecology's opinion/detailed rules regarding preimplantation genetic testing for infertility and recurrent pregnancy loss, and is intended for the following patients who have been confirmed at an ART facility (medical institution).
・Infertile couples with a history of repeated failed IVF embryo transfer attempts.
・A couple with recurrent pregnancy loss who has a history of repeated miscarriages and stillbirths.
・A couple with recurrent pregnancy loss(or infertility) in which one of the partners has been confirmed to have a chromosomal structural abnormality (such as a balanced chromosomal translocation). However, there is no question as to whether or not a patient has a history of pregnancy or miscarriage/ stillbirth.

There are two types of barcode labels: test ID stickers and specimen ID stickers. Please attach the test ID sticker to the PGT-A tube box (paper box) and the request form. Please attach the specimen ID stickers to the required number of containers to be submitted, wrapping them around the top. Also, please attach the corresponding specimen ID sticker to the request form.
Please add 2.5μL of PGT-A specimen preservation solution to the submission container beforehand. After adding, spin down to make sure the preservation solution is at the bottom of the PCR tube.
Please collect 5-10 trophectoderm cells from the cultured fertilized egg (blastocyst) and place them in the submission container.
Please take the following precautions into consideration in order to collect better quality specimens.
1. Be sure to wear gloves to prevent contamination with workers' cells.
 2. Collect the specimen from the collection drop with a micropipette, wash it by pipetting multiple times (approximately six times) in the washing drop, and then inject it into the 2.5 μL preservation solution drop on the bottom of the submission container mentioned above under a microscope (by doing this under a microscope, make sure that the collected specimen has been placed securely in the storage container).
 3. Immediately spin down to ensure the specimen is at the bottom of the tube.
Please place the collected submission container on the PGT-A tube box stand (24 holes). Place them in a PGT-A tube box with the number of tubes written on it in permanent marker. Store the PGT-A tube box frozen (-20℃ or lower). After freezing, do not allow the tube to freeze and thaw.
