Radioimmunoassay
A method to perform the competitive antigen-antibody reaction between the antigen labeled with radioisotope (RI) against the antibody and the antigen in the specimen, and to separate the labeled antigen bound to the antibody (bound type: Bound) from the labeled antigen not bound to the antibody (free type: Free) and to measure the ratio of them as the antigen concentration from the radioactivity.
As separation methods for bound and free types (B/F separation), there are the solid-phase method in which the antibody is solid-phased, the two-antibody method in which a second antibody is bound to the antigen-antibody complex and precipitated, the ammonium sulfate precipitation method in which the antigen-antibody complex is precipitated with ammonium sulfate (ammonium sulfate), and the PEG method in which the antigen-antibody complex is precipitated with a precipitation reagent.
